Divergence of ovarian follicular growth response in broiler breeder hens provided twice the recommended amount of feed

PL10 enhanced fusion (82.4 ± 1.4 vs. 58.4 ± 3.0% sperm fused, PL10 vs. PL20, n = 7; P < 0.0001) and sperm viability (27 ± 2, 46 ± 3, and 39 ± 4%, respectively, for PL, PL10, and PL20, P < 0.05). Because viability was still unacceptably low, we then added poly-L-lysine to the PL10 recipe at 1 (PL10−1), 5 (PL10−5), and 10 mol% (PL10−10). Significantly more sperm fused to PL10−1 than PL10 (93.8 ± 0.8 vs. 79.1 ± 4.8%, n=4, P < 0.05); fusion to PL10−5 and PL10−10 was intermediate. Motility and viability were determined for sperm alone (CON) and sperm fused to PL10 and PL10−1 at 1, 30, 60, 120, 180, and 240 minutes. As expected, motility decreased steadily over time for all treatments. However, motility of PL10-treated sperm was significantly lower than other sperm from 30 minutes on. CON and PL10−1 sperm had similar motilities until 240 min, when PL10−1 was better (4 ± 2 vs. 16 ± 8%, respectively, for CON and PL10−1, P < 0.05). Viability was improved by PL10−1 and PL10 compared to CON (P < 0.05). Fusing proteoliposomes to avian sperm appears to improve sperm function.
